Fault code associated with O2-1, O2-2, Intake air pressure sensor, coolant temperature sensor, throttle position sensor, and many more sensors feedback error, is cause and Cruise control system cannot be operated before issue is fixed.
In your case it looks like after ECU flash/tuning it stopped mostly because of some glitch in program related to O2 sensor. Recommend to get corrected program from tuner.
Tips: in some case, even thou if you disable O2 sensor, it some how interfere with tuned ECU if electrically connected or not removed from motorcycle. Completely isolate O2 sensor from ECU after disabling it on ECU.

You got the diagnostic tool, did it show ANY engine codes? If so, which ones?

On the 2022+ MT10, both Oxygen sensors require being connected for the cruise control to operate, whether it be tuned or not. There are 2 sensors, one of the connectors is located on the LH side of the engine, which is easily accessible. The other connector requires lifting the fuel tank for access. IF both plugs are disconnected, the cruise light will flash and not operate. If the sensor plug located on the LH side of the engine is connected and the one under the tank is disconnected, the cruise light will illuminate solid, but still not operate. BOTH plugs require being connected for cruise to operate. The Tuner should've put the O2 sensors into closed loop. The tune done on mine here in Australia gave me 155hp (140hp factory) at the wheels and 108Nm (98Nm factory).
